However, departures from usual seasonal trends are often reported. To understand the role of local stressors in modifying seasonal succession, we sampled fortnightly, throughout three seasons, five nearby shallow soda lakes exposed to identical seasonal and meteorological changes. We characterised their microeukaryotic and bacterial communities by amplicon sequencing of the 16S and 18S rRNA gene, respectively. Biological interactions were inferred by analyses of synchronous and time-shifted interaction networks, and the keystone taxa of the communities were topologically identified. The lakes showed similar succession patterns during the study period with spring being characterised by the relevance of trophic interactions and a certain level of community stability followed by a more dynamic and variable summer-autumn period. Adaptation to general seasonal changes happened through shared core microbiome of the lakes. Stochastic events such as desiccation disrupted common network attributes and introduced shifts from the prevalent seasonal trajectory. Its salinity varies within a wide range (from 6.3 to > 72 parts per thousand). Despite these extreme conditions, phytoplankton blooms are often observed. Therefore, our aim was to study the key abiotic drivers and ecological optimum of the picoplankton in a 5-year period (2013-2017). The taxonomic composition, long-term dynamics and abiotic drivers of the autotrophic picoplankton (APP) has been investigated in North Saltern of the coastal lagoon in Atanasovsko Lake. The APP species were identified by using a polyphasic approach based on molecular and phylogenetic analyses of 16S/18S rDNA in combination with cytomorphological and ecological characterization. Picochlorum oklahomense (Trebouxiophyceae, Chlorophyta) and marine Synechococcus sp. (Synechococcales, Cyanobacteria) were the dominant genotypes within the APP. The relative biovolume of the APP to the total phytoplankton biovolume was strongly variable (from 36 to 99%) as the salinity and temperature were found to be the main abiotic drivers of its dynamics. Salinity between 30-59 parts per thousand and mid-summer temperature above 24 degrees C provide an ecological optimum for the APP development at a relative biovolume of 93%. The area is characterised by World War II bomb crater ponds and can be regarded as an excellent ecological model system where the dispersion of species is slightly limited by distance. To study the effect of environmental variables on the communities, canonical correspondence analysis was applied. Salinity, pH, total suspended solids, total phosphorous and depth proved to be significant environmental drivers in this analysis. The ecological status of the ponds was assessed with Ziemann's halobity index, as the trophity-depending metric cannot be applied to these habitats (due to the naturally high phosphorus content). Ponds in "good" ecological status significantly differed from those appertaining to water quality category of "not-good" ecological status considering characteristic of natural astatic soda pans (e.g. salinity, pH, ammonium, total phosphorous concentration, nitrogen:phosphorous ratio and turbidity). The differences between epiphytic diatom communities inhabiting the ponds were detected using non-parametric multidimensional scaling. The samples formed three groups according to the types of ponds ("transparent", "transitional" and "turbid") based on the width of the macrophyte belt around them. Indicator species related to the ecological status of the ponds and diatom communities contributing to the separation of groups of ponds were identified. One of the indicator species differed from species already described. Light and scanning electron microscopy features and phylogenetic analyses based on three genes (18S and 28S rRNA genes, rbcL) proved that it was a new species of Nitzschia genus, closely related to Nitzschia frustulum and Nitzschia inconspicua. Therefore, description of a new species, Nitzschia reskoi Acs, Duleba, C.E.Wetzel & Ector is proposed. Показано, что ни один критерий в отдельности: морфологический или иной признак, вычисление генетических дистанций, анализ и сравнение вторичной структуры ITS2, поиск компенсаторных замен и молекулярных подписей - не позволяет надежно классифицировать таксоны. В связи с чем необходимо использовать полифазный подход в систематике водорослей, особенно при идентификации таксонов с простой клеточной морфологией, которые часто встречаются в почвах.The article describes the morphological and molecular genetic approaches used to identify the green microalgae of the Trebouxiophyceae class from the ACSSI collection. In the brown semi-desert and chestnut soils, we found Muriella terrestris. In the buried meadow-chestnut soil we found Edaphochlorella mirabilis. Three more strains are characterized as new uncharacterized taxa: ACSSI 014 is a new species related to the genus Watanabea isolated from gray forest soil, with a pyrenoid surrounded by several starch grains, in contrast to the type species, and ACSSI 104 and 144 are a Nannochloris -like genus, whose representatives were found in solonetz and chestnut soil and are characterized by poor morphological specificities. It is shown that no single criterion (either morphological or other specificity, calculation of genetic distances, analysis and comparison of the ITS2 secondary structure, search for compensatory changes and molecular signatures) does not allow reliable classification of taxa. The pans are highly productive environments with picophytoplankton predominance. Little is known about the planktonic bacterial communities inhabiting these aquatic habitats; therefore, amplicon sequencing and shotgun metagenomics were applied to reveal their composition and functional properties. Results showed a taxonomically complex bacterial community which was distinct from other soda lakes regarding its composition, e.g. the dominance of class Alphaproteobacteria was observed within phylum Proteobacteria. The shotgun metagenomic analysis revealed several functional gene components related to the harsh and at the same time hypertrophic environmental conditions, e.g. proteins involved in stress response, transport and hydrolase systems targeting phytoplankton-derived organic matter.
