"Our present review is focusing on the uniqueness of balanced \nastroglial signaling. The balance of excitatory and inhibitory \nsignaling within the CNS is mainly determined by sharp synaptic \ntransients of excitatory glutamate (Glu) and inhibitory gamma-\naminobutyrate (GABA) acting on the sub-second timescale. \nAstroglia is involved in excitatory chemical transmission by \ntaking up i) Glu through neurotransmitter-sodium transporters, \nii) K+ released due to presynaptic action potential generation, \nand iii) water keeping osmotic pressure. Glu uptake-coupled Na+ \ninflux may either ignite long-range astroglial Ca2+ transients \nor locally counteract over-excitation via astroglial GABA \nrelease and increased tonic inhibition. Imbalance of excitatory \nand inhibitory drives is associated with a number of disease \nconditions, including prevalent traumatic and ischaemic \ninjuries or the emergence of epilepsy. Therefore, when \naddressing the potential of early therapeutic intervention, \nastroglial signaling functions combating progress of Glu \nexcitotoxicity is of critical importance. We suggest, that \nexcitotoxicity is linked primarily to over-excitation induced by \nthe impairment of astroglial Glu uptake and/or GABA release. \nWithin this framework, we discuss the acute alterations of Glu-\ncycling and metabolism and conjecture the therapeutic promise of \nregulation. We also confer the role played by key carrier \nproteins and enzymes as well as their interplay at the \nmolecular, cellular, and organ levels.
