"Vinpocetine (ethyl-apovincaminate, Cavinton), a synthetic \nderivative of the Vinca minor alkaloid vincamine, has been used \nnow for decades for prevention and treatment of cerebrovascular \ndiseases predisposing to development of dementia. Both \nvinpocetine and its main metabolite cis-apovincaminic acid \n(cAVA) exert a neuroprotective type of action. Bilateral N-\nmethyl-D-aspartate (NMDA)-induced neurodegeneration in the \nentorhinal cortex of rat was used as a dementia model to confirm \nthe neuroprotective action of these compounds in vivo. NMDA-\nlesioned rats were treated 60 min before lesion and throughout 3 \npostoperative days with a 10 mg/kg intraperitoneal dose of \nvinpocetine or cAVA. Behavioral tests started after termination \nof drug treatment and consisted of novel object recognition, \nsocial discrimination, and spontaneous alternation in a Y-maze, \nand spatial learning in the Morris water maze. At the end of \nbehavioral testing brains were perfused with fixative and the \nsize of the excitotoxic neuronal lesion and that of microglial \nactivation around the lesion were assayed quantitatively on \nbrain sections immunostained for neuron-specific nuclear protein \n(NeuN) and integrin CD11b, respectively. Entorhinal NMDA lesions \nimpaired recognition of novel objects and the new social \npartner, and suppressed spontaneous alternation and spatial \nlearning performance in the Morris maze. Both vinpocetine and \ncAVA effectively attenuated the behavioral deficits, and \nsignificantly decreased lesion size and the region of microglia \nactivation. Both lesion-induced attention deficit and learning \ndisabilities were markedly alleviated by vinpocetine and cAVA. \nThe morphological findings corroborated the behavioral \nobservations and indicated reduced lesion size and microglia \nactivation especially after vinpocetine treatment which supports \nan in vivo neuroprotective mode of action of vinpocitine and a \nless potent action of
