AB - In this study, we report the synthesis and detailed characterization of four novel bidentate (N, N) ligands incorporating a 2-(methylthio)pyrimidine moiety and their fac-tricarbonylrhenium(I) complexes (ReB1—ReB4 and ReB1Aq) with the general formula fac-[Re(CO)3(N, N)X]n+, with X = Cl—or H2O and n = 0 or 1. Designed to integrate biologically relevant functionalities, these complexes exhibited promising multifunctional bioactivity. Cytotoxicity assays demonstrated moderate activity (IC50 = 11–78 µM) on various human cancer cell lines, with certain derivatives showing notable selectivity toward the Colo205 line. Most of the chlorido complexes effectively inhibited the replication of Herpes simplex virus type 2, while ReB4 displayed significant antibacterial activity against Staphylococcus aureus, including methicillin-resistant strains (MIC = 12.5–25 µM), and demonstrated biofilm inhibition. Aqueous stability of these organometallic complexes was thoroughly investigated, and complexes ReB2 and ReB3 containing a pyrimidine and a thiazole ring, respectively, gradually decompose in aqueous media, correlating with a decline in anticancer activity. Ligand-exchange processes were observed, in which the chlorido co-ligands were replaced by water, thus affecting the solubility and lipophilicity. The aqua complex ReB1Aq exhibited a low chloride affinity, and the pKa of the coordinated water molecule was obtained to be ∼8. Its interaction with human serum albumin was investigated in detail and was found to be dominated by non-covalent interactions, indicating that no coordination bond formation occurs with the protein. Vernoniastrum migeodii (S. Moore) Isawumi (Asteraceae) represents a chemically promising but understudied source of bioactive small molecules. This study aimed to define the metabolite profile of V. migeodii and evaluate the modulation of inflammatory epithelial signaling of the constituents. From the methanolic extract of V. migeodii, five germacranolide sesquiterpenes, vernolide (1), 3′-hydroxylvernolide (2), pectorolide (3), 4′-hydroxypectorolide-14-O-acetate (4) and 4′-hydroxypectorolide (5), together with (6S,9R)-vomifoliol (6), eucarvone (7), luteolin (8), and luteolin-7-O-glucoside (9) were isolated by multiple chromatographic separations. The structures were determined by comprehensive 1D and 2D NMR spectroscopy. Isolated compounds 1 to 9 together with previously reported steroids (10–17) and tripeptide (18) were evaluated in LPS-activated A549 cells by quantitative PCR for interleukin-6 (IL6), interleukin-1β (IL1β), and prostaglandin-endoperoxide synthase 2 (PTGS2) and by enzyme-linked immunosorbent assay (ELISA) for IL-6 and IL-8. Compounds 2, 7, steroids 10–17 and aurantiamide acetate (18) reduced IL6 mRNA relative to the LPS control, while (6S,9R)-vomifoliol (6) increased IL-6 and elevated IL-8. In the assay IL1β and PTGS2 transcripts were not significantly altered. These findings highlight the potential of V. migeodii metabolites as modulators of epithelial inflammatory pathways. Combining chemical and biological evidence provides a clear basis for structure–activity- and pathway-focused studies. Hybridizing these pharmacophores and further modifying them via modified Mannich reactions provides a strategy to improve their physicochemical parameters and selectivity toward multidrug-resistant (MDR) cancer cells. Objectives: To synthesize a series of 8-hydroxyquinoline–phthalimide hybrids and their Mannich base derivatives and evaluate their cytotoxic activity and resistance-selective properties against sensitive Colo205 and resistant Colo320 cancer cell lines. Methods: Four hybrid compounds were synthesized by reacting 5-amino-8-hydroxyquinoline with different phthalic anhydride derivatives. Twelve fine-tuned derivatives were prepared by using the modified Mannich reaction. Cytotoxic activity was measured using the MTT assay, and relative resistance (RR) was calculated to determine selectivity toward the resistant cell line. P-glycoprotein (Pgp) ATPase activity was evaluated for the most active compounds. Results: All derivatives displayed cytotoxic activity, with higher potency toward the resistant Colo320 cell line. Compounds 2 and 4 showed the strongest activity against both cell lines (IC50 down to 4.88 µM). Compounds 5, 8a, 9a, and 9c retained potent activity against Colo320 (IC50 = 9.89–22.79 µM). Incorporating a CH2–N group at position C7 substantially enhanced the selectivity for MDR cells. Compounds 9c, 9a, and 8a exhibited the highest selectivity, with RR values of 0.29, 0.33, and 0.35, respectively. Compounds 2, 4, 5, 8a, and 9a showed inhibitory effects on Pgp ATPase activity. Conclusion: The newly synthesized HQ–phthalimide hybrids represent promising candidates for targeting MDR in colorectal cancer, with Mannich modification enhancing the selectivity toward resistant cells. This narrative review explores the diversity of beta-lactamases found in intestinal anaerobes, their functional characteristics, and clinical implications. Species of the genus Bacteroides are major producers, harboring enzymes from Ambler classes A (e.g. CepA, CfxA and CblA), B (e.g. CfiA and CcrA) and D (e.g. OXA-347), with activity ranging from cephalosporinases to carbapenemases and oxacillinases. Other gut members, including Clostridioides difficile, Fusobacterium nucleatum, and Acidaminococcus fermentans, also encode specific beta-lactamases. These enzymes contribute to microbiota resilience following beta-lactam exposure and can protect neighboring bacteria via extracellular degradation. This protective mechanism has inspired therapeutic applications, such as recombinant beta-lactamases (ribaxamase, SYN-006) designed to preserve gut integrity during antibiotic therapy. Understanding these intrinsic resistance mechanisms is essential for optimizing antibiotic stewardship and mitigating the spread of resistance genes. Copyright © 2026. Published by Elsevier Ltd.
