AB - Cyclodextrin polymers (CDPs) are a class of polymers that have been extensively used in diverse scientific and technological fields. These polymers are characterized by the presence of cyclodextrins (CDs) as repeating units within their polymeric networks. The unique structure of CDs has enabled the synthesis of CDPs using multifunctional reagents, resulting in an enormous variety of CDPs with diverse structures and properties. This review article focuses on a key subclass of CDPs, namely those containing ionizable and ionic groups (I-CDPs). These polymers can be synthesized by introducing ionizable and ionic groups into the CDPs networks. The first I-CDPs were introduced in 1985, and, since then, there have been significant advances in their synthesis methods and applications. The remarkable features of I-CDPs are their pH-tunable water solubility and high and tunable binding affinity toward various guests, which are difficult to achieve by charge-neutral CDPs. In this review, we categorize I-CDPs into cationic, anionic, and ampholytic polymers and provide an overview of the progress made in their synthesis and applications. We highlight the creative methods used to introduce charged groups into the structure of CDPs and discuss the properties of I-CDPs that make them suitable for diverse applications. © 2024 Elsevier Ltd

AB - The first continuous flow method was developed for the synthesis of 6-monoamino-6-monodeoxy-β-cyclodextrin starting from native β-cyclodextrin through three reaction steps, such as monotosylation, azidation and reduction. All reaction steps were studied separately and optimized under continuous flow conditions. After the optimization, the reaction steps were coupled in a semi-continuous flow system, since a solvent exchange had to be performed after the tosylation. However, the azidation and the reduction steps were compatible to be coupled in one flow system obtaining 6-monoamino-6-monodeoxy-β-cyclodextrin in a high yield. Our flow method developed is safer and faster than the batch approaches.

AB - Highly resistant bacteria producing metallo-beta-lactamases (MBLs) to evade beta-lactam antibiotics, constitute a major cause of life-threatening infections world-wide. MBLs exert their hydrolytic action via Zn2+ cations in their active center. Presently, there are no approved drugs to target MBLs and combat the associated antimicrobial resistance (AMR). Towards this issue, we have prepared a family of cyclodextrins substituted with iminodiacetic acid (IDA) on their narrow side, while the wider side is either unmodified or per-2,3-O-methylated. The molecules form strong coordination complexes with Zn2+ or Ga3+ cations in aqueous solution. Free and metal-complexed compounds have been thoroughly characterized regarding structures, pH-dependent ionization states, distribution of species in solution, pKa values and metal-binding constants. At neutral pH the multi-anionic hosts bind up to four Zn2+ or Ga3+ cations. In vitro, 50 mu M of the compounds achieve complete re-sensitization of MBL-producing Gram-negative clinical bacterial strains resistant to the carbapenems imipenem and meropenem. Moreover, the radioactive complex [Ga-67]Ga-beta-IDACYD prepared, displays high radiochemical purity, sufficient stability both overtime and in the presence of human plasma apo-transferrin, thus providing an invaluable tool for future biodistribution and pharmacokinetic studies of beta-IDACYD in vivo, prerequisites for the development of therapeutic protocols.
