It has been reported that the lack of CypD affects glucose and lipid metabolism. However, the findings are controversial regarding the metabolic pathways involved, and most reports describe the effect of a high-fat diet on metabolism. We performed a lipidomic analysis of plasma and liver samples of CypD-/- and wild-type (WT) mice to reveal the lipid-specific alterations resulting from the absence of CypD. In the CypD-/- mice compared to the WT animals, we found a significant change in 52% and 47% of the measured 225 and 201 lipid species in liver and plasma samples, respectively. The higher total lipid content detected in these tissues was not accompanied by abdominal fat accumulation assessed by nuclear magnetic resonance imaging. We also documented characteristic changes in the lipid composition of the liver and plasma as a result of CypD ablation with the relative increase in polyunsaturated membrane lipid species. In addition, we did not observe remarkable differences in the lipid distribution of hepatocytes using histochemistry, but we found characteristic changes in the hepatocyte ultrastructure in CypD-/- animals using electron microscopy. Our results highlight the possible long-term effects of CypD inhibition as a novel therapeutic consideration for various diseases. The discrimination of the geometric isomers of fatty acid needs proper and effective separation conditions. The efficiency of three different stationary phases was evaluated by GC–MS methods in the separation of fatty acids in their methyl ester forms. Significant differences were observed in the efficiencies of polysiloxane-based (non-polar HP-5MS and medium/high polarity DB-225MS) and ionic liquid-based (SLB-IL111) columns. Baseline separation of the geometric isomers of linoleic acid methyl ester was obtained by the extremely polar SLB-IL111 column, showing a preference over the other two columns. The optimization of the experimental conditions (response linearity, limit of detection, limit of quantification, system suitability, intraday and interday repeatability and accuracy) showed the separation power of the ionic liquid interaction in the analyses by using short (25–30 m long) columns. By deducting the general principles of the interaction, predictions can be made for the separation of other isomers. The results facilitate the precise identification of various types of fatty acids in real samples for nutritional information. According to recent studies heated and several times heated oils may play a role in the pathophysiology of certain chronic diseases. In our study we investigated the effects of repeated heating on the fatty acid composition of plant-based cooking oils Materials and methods: We investigated five, in the household commonly used plant-based oils and fats. Potatoes were fried on 180 oC in sunflower-, rapeseed-, soy-, palm- and extra virgin olive oil and then we let the oils cool down to 70 oC. After sieving this procedure was repeated ten times. Samples were collected from each fresh, unheated oil as well as after the 1st, 5th and 10th frying period. Fatty acid composition of the samples was determined by Agilent 6890 GC with COC and FID. Chromatograms were evaluated with Chromeleon 7.2 software, for statistical analysis SPSS 25.0 program was used for ANOVA tests and Post Hoc analysis. Results: Values of both essential fatty acids: n-3 alpha-linolenic acid and n-6 linoleic acid were significantly decreased in the course of the heating period, while values of the investigated trans isomers as well as the sum of trans isomers significantly increased except for soy oil. Both the calculated iodine value and the unsaturated to saturated fatty acid ratio significantly decreased in all of the investigated oils.
