AB - Vascular graft infection is still a life-threatening complication of reconstructive surgery. Among other options, application of cryopreserved homografts can eventuate favorable outcome, if graft replacement is necessary. The preparation and storage of these allografts need special infrastructure and deep subzero (− 80 °C) temperature. However, the longer storage time can lead to inferior results after implantation, based upon clinicians’ experiences. The goal of our investigation was to circumscribe the optimal storage time interval with differential scanning calorimetry (DSC) and histological evaluation, using porcine aorta. All samples were deep-freezed using − 80 °C. Cryopreservated grafts were melted after 4, 6, 12, 16, 20, 24, 28 and 52 weeks; then, DSC and different types of histology were performed. Light microscopy analysis showed significant changes in the connective tissue fibers’ structure from the 16th week; while, DSC measurements confirmed systematic decrease in the thermal stability from the same week during the follow-up period. Our investigation suggested that cryopreservation can lead to significant and increasing microstructural damage of the fibers following the 12th week; thus, the homograft implantation can result in higher success rate inside this timeframe. The gelsolin (GSN) regulates the remodeling of cytoskeleton. After the activation of GSN by calcium ions, it can sever actin filaments then capped at its barbed end. In the cytoplasm, GSN manages the cellular motions and morphology. Phosphatidylinositol 4,5-bisphosphate (PIP2) is involved in signal transduction and the regulation of the actin cytoskeleton by regulation of actin-binding proteins. GSN can bind to PIP2 and thus can be localized in the near of the plasma membrane and released from the end of F-actin. We test here with isoperibol calorimetry the enthalpy change, within the interplay between GSN and F-actin under nano-, micro- and millimolar calcium concentrations and express the importance of PIP2 binding for the inactivation of GSN. As we have demonstrated here that PIP2 binding stabilizes the structure of gelsolin and reduces its actin monomer binding activity under nanomolar calcium as the typical cytoplasmic calcium concentration of resting cells. The gelsolin shows partial activity under micromolar and total activity with strong responses under millimolar calcium. If gelsolin-capped filaments point at the plasma membrane helps the binding between gelsolin and PIP2, and hence, filament uncapping in case of resting cells. We presume that the low free calcium concentration keeps on the structure of gelsolin which is able to bind actin within the cooperativity of actin bound calcium. Gelsolin can help to manage monomer pool far from the membrane and it can be linked to a basic sensory mechanism which drives the direction of filament growth in the near of the membrane. Leiomodins 1–3 (Lmod1–3) are crucial for the development of muscle sarcomeres. Unlike tropomodulins that localize only at the pointed ends, the striated muscle specific Lmod2 shows diffuse distribution along the entire length of the thin filaments. The G-actin-binding profilin (Pro) facilitates the nucleotide exchange on monomeric actin and inhibits the polymerization at the barbed end, therefore contributes to the maintenance of the intracellular pool of polymerization competent ATP-G-actin. Cyclophosphamide (CP) is a cytostatic drug that can have potential side effects on muscle thin filaments at the level of actin in myofilaments. Here, we aimed at investigating the influence of CP on actin and its complexes with actin-binding proteins by using differential scanning calorimetry (DSC). We found that upon CP treatment, the denaturation of the Pro-G-actin and Lmod2-F-actin complexes was characterized by an increased enthalpy change. However, after the CP treatment, the melting temperature of F-actin was the same as in the presence of Lmod2, seems like Lmod2 does not have any effect on the structure of the CP alkylated F-actin. In case of Pro bound G-actin the melting temperature did not respond to the CP addition. The intracellular function of Lmod2 in muscle cells can be modified within CP drug treatment. From this survey, the third most common is breast cancer, the fifth is melanoma malignum and pancreatic adenocarcinoma ranks twentieth. Undoubtedly, the early diagnosis and monitoring of these tumors and related research is important for aspects of patient care. The aim of our present review was to explain an impressive methodology that is deemed suitable in reference to studying blood sample deviations in the case of solid tumors. Essentially, we compared the heat denaturation responses of blood plasma components through differential scanning calorimetry (DSC). In the control, between five and seven separable components can be detected, in which the primary component was albumin, while in the case of tumorous patients, the peaks of immunoglobulins were dominant. Moreover, the shape of the plasma DSC curves changed with a shift in the higher temperature ranges; thus, their pattern can be used as a suitable marker of direct immunological responses. The further development of the analysis of DSC curves raises the possibility of the early diagnosis of a potential tumor, the monitoring of diseases, or testing the efficacy of the therapy from a single drop of blood. Despite the recent advances in imaging technologies, there is still a need for more information about the degree of joint degeneration and changes of the collagen content of tendons to select the proper implant type. It is known that human long head biceps tendon subjected to synovial inflammation and mechanical wear could result in tear and structural damage. Differential scanning calorimetry has already been demonstrated to be a dependable in the assessment of collagen in degenerative tendon conditions. The purpose of the study was to determine whether variations in thermal parameters detected by DSC are related to the degenerative changes of LHBT samples. LHBT tendons were collected from patients who had reversed shoulder arthroplasty because of trauma. DSC was utilized to measure the thermodynamic changes. Macroscopical, radiological and histological assessment were also performed. Based on the thermal parameters and denaturation curves, LHBT samples collected of patients underwent reversed shoulder arthroplasty due to comminuted proximal humerus fracture have shown minimal or mild degenerative injuries. These data were supported by preoperative radiological examination and histological analysis. The authors suggest that DSC could be a useful investigative tool in the clinical evaluation of tendon injuries. LA - English DB - MTMT ER -
