A promising modern agriculture method is the foliar treatment of zinc oxide nanoparticles (ZnONPs). This approach has shown promise in boosting challenged tomato yields, fruit quality, and leaf extract antibacterial activity against pathogens. A greenhouse experiment was conducted. The previously synthesized and characterized ZnONPs were used to alleviate the harmful effects of NaCl stress. Tomato fruit weight from different treatments was determined, and the gas–liquid chromatography device was used to observe the changes in fatty acid production. The antimicrobial activities of the aqueous and diethyl ether extracts from tomato leaves were determined against six bacterial and six fungal strains. The plants that were salinity-stressed and sprayed with 0.075 and 0.15 g/L ZnONPs showed a better improvement compared to the salinity-stressed plants. Also, the sprayed plants that were not stressed at all showed promising results compared to the control and the other different treatments. Through the process of molecular docking, it was shown that caffeic acid, ferulic acid, p-coumaric acid, sinapic acid, and apigenin-7-glucoside are essential chemicals that possess antibacterial and antifungal effects against the DNA Gyrase inhibitor and the sterol 14-alpha demethylase (CYP51) enzyme, respectively. It is concluded that salt stress can negatively affect the growth, quality, and variant plant features. However, the foliar application of ZnONPs is able to overcome those adverse effects in the stressed plants, and enhance the non-stressed as well. It requires an investigation of ameliorative techniques, for example, spraying synthesized nanoparticles such as zinc oxide nanoparticles (ZnOnps). This current research studied the impact of sodium chloride as a stressor (150 mM NaCl) and the application of ZnOnps (2 g L−1) on some biochemical properties of maize (Zea mays) leaves. The experiment involved examining some mineral concentrations (Na, K, Mg, Zn, Cu, Mn), fatty acid profile, and the antimicrobial (antibacterial and antifungal) properties of aqueous and diethyl ether maize leaf extracts, supported by molecular docking studies of the 17 previously determined phenolic compounds against DNA gyrase and alpha-L-fucosidase enzymes. Applying ZnOnps markedly decreased sodium concentrations from 5.8 to 1.9 mg g−1 dry weight (DW) and established ion balance. ZnOnps also reduced γ-linolenic acid levels to 60% under stress, returning them to normal (34%), while increasing palmitic acid to 30%. Determining the antimicrobial activities indicated that extracts from plants sprayed with ZnOnps exhibited enhanced antimicrobial activity, as evidenced by the lowest minimum inhibitory concentrations against bacterial and fungal strains, including Salmonella typhi and Aspergillus flavus. The computational molecular docking confirmed the antimicrobial findings, with the compound apigenin-7-glucoside, which exhibited the highest binding affinity scores for antibacterial (−7.4 kcal/mol), and the compound chlorogenic acid as antifungal (−7.2 kcal/mol) against the enzyme targets. Thus, ZnOnps can be considered an efficient strategy for mitigating salinity stress in maize plants while elevating the antimicrobial activity and stability of variant secondary compounds. Transmission and scanning electron microscopy (TEM and SEM), energy-dispersive X-ray spectroscopy (EDAX), and X-ray diffraction confirmed the nanoparticles’ hexagonal morphology and typical dimensions of 51 nm. The size and stability of these nanoparticles were assessed through the size distribution and zeta potential analysis, indicating reasonable stability. Fourier-transform infrared spectroscopy (FTIR) detected the newly formed functional groups. This study emphasized the role of reactive oxygen species (ROS) and phenolic compounds in plant responses to nanoparticle treatment, particularly in detoxifying harmful radicals. The research also examined the activity of antioxidant enzymes, including peroxidase (POX), catalase (CAT), and glutathione reductase (GR), in alleviating stress caused by oxidation while subjected to various treatments, including micronutrient seed priming with DR GREEN fertilizer. Some biochemical compounds, such as total phenolics (TPCs), total flavonoids (TFCs), and total hydrolysable sugars, were estimated as well to show the effect of the different treatments on the wheat plants. The findings suggested that ZnO nanoparticles can enhance antioxidant enzyme activity under certain conditions while posing phytotoxic risks, underscoring the complexity of plant–nanoparticle interactions and the potential for improving crop resilience through targeted micronutrient applications. Seed-priming treatment was applied to winter wheat genotype AG Hurrem with Dr. Green seed-priming fertilizer, which is a commonly used seed fertilizer containing macro- and microelements. Genome-wide transcriptomic analysis was performed to examine the effects of treatments. In seed-primed plants, defense response pathways such as purine and thiamine metabolism, glutathione pathway, and phenylpropanoid biosynthesis were activated. At the same time, photosynthesis and some cellular respiration processes were downregulated and suppressed. Furthermore, in samples of plants previously exposed to priming and subsequently to drought stress, biochemical pathways activated during seed priming showed positive modulation, thus confirming the long-term traces of the priming effects of previous treatments and their repeated inducibility in the genome, i.e., the presumed existence of stress memory. The in silico analyses were also supported by laboratory antioxidant enzyme activity measurements. The priming technique and the preventive approach that can be offered with it may be a promising option for developing sustainable agricultural production in the future.
