The black mulberry tree, Morus nigra L.(Moraceae), is known for its very high total phenolic and flavonoid content, making it the richest source of such compounds when compared to other Morus species. The root bark of M. nigra has been reported to possess a broad range of bioactivities including antihypertensive effect, supporting its related traditional use2. Nevertheless, only limited information is available on the ACE inhibitory activity of this drug or its constituents. In the present study, a methanolic extract of M. nigra root bark was subjected to a phytochemical study through the consecutive use of various preparative chromatographic techniques, leading to the isolation and characterization of seven phenolic compounds. Their chemical structures were elucidated by one- and two-dimensional NMR spectroscopy and HRMS, identifying them as prenylated arylbenzofurans such as Moracin P (1) and Moracin R (2), and prenylated flavonoids such as Kuwanon U (3), Morusin (4), Kuwanon E (5), Albanol A (6) and Albanol B (7). Compounds 1-7 were evaluated for their potential to inhibit ACE in vitro. Our preliminary results revealed the highest ACE inhibition activity for compounds 5- 7, each showing a complete inhibition of the enzyme at a concentration of 36.4 μg/mL, whereas compound (3) exhibited nearly 50% inhibition at the same concentration. Our results suggest that M. nigra is an excellent source of potentially anti-hypertensive compounds, and that ACE inhibition by its prenylated phenolic compounds might provide a rationale for its related traditional use.
