вторичных заболеваний таких, как нефропатия, ретинопатия, нейропатия и ангиопатия. Различные гликирующие карбонильные соединения, например, 3-дезоксиглюкозон (3-ДГ), образующиеся при патологиях, приводят к накоплению больших количеств ППГ in vivo и in vitro. В своей предыдущей работе мы осуществили гликирование гистона H2A под действием 3-ДГ и наблюдали образование различных промежуточных и конечных продуктов гликирования, а также изменения структуры молекулы белка. В представленной работе изучены иммуногенные свойства нативного и 3-ДГ-модифицированного гистона H2A на самках кроликов. Показано, что модифицированный H2A оказался высоко иммуногенным и вызывал образование специфических антител в высоком титре, тогда как нативный белок почти не стимулировал продукцию специфических иммуноглобулинов. В крови кроликов, иммунизированных гликированным H2A, были обнаружены антитела к стандартному карбоксиметиллизину (КМЛ) и пентозидину, что свидетельствует о высокой иммуногенности этих природных ППГ. Полученные результаты свидетельствуют о том, что ППГ и структурные нарушения белка обладают способностью активировать иммунную систему благодаря формированию в белковой молекуле новых антигенных детерминант, придающих ей высокую иммуногенность. В своем исследовании мы обнаружили присутствие аутоантител против 3-ДГ-модифицированного H2A, КМЛ и пентозидина в сыворотке больных диабетом 2-го типа, осложненным вторичными заболеваниями. Исследования уровня аутоантител к поврежденному H2A и ППГ могут быть важны при оценке начала или прогрессирования вторичных заболеваний, возникающих у пациентов с сахарным диабетом 2-го типа. Кроме того, эти антитела могут использоваться в качестве маркеров для ранней диагностики вторичных заболеваний, осложняющих диабет.Advanced glycation endproducts (AGEs) have been found to be critically involved in initiation or progression of diabetes secondary complications (nephropathy, retinopathy, neuropathy, and angiopathy). Various hyperglycating car-bonyl compounds such as 3-deoxyglucosone (3-DG) are produced in pathophysiological conditions that form AGEs in high quantity both in vivo and in vitro. In the first stage of the study, we glycated histone H2A protein by 3-DG, and the results showed the formation of various intermediates and AGEs as well as causing structural changes in the protein. In the second stage, the study demonstrated the immunogenicity of native and 3-DG glycated H2A protein in female rabbits. The modified H2A was highly immunogenic, eliciting high-titer immunogen-specific antibodies, while the unmodified form was almost non-immunogenic. Antibodies against standard CML and pentosidine were detected in the immunized female rabbits, which shows the immunogenic nature of AGEs (CML and pentosidine) as well. The results show both structural perturbation and AGEs have the capacity of triggering the immune system due to the generation of neoepitopes that render the molecule immunogenic. This study shows the presence of autoantibodies against 3-DG modified H2A, CML (carboxymethyllysine), and pentosidine in the sera of type 2 diabetes patients having secondary complications.
